摘  要:动用全外实验,对NADPH/FeSO4诱发的微粒体脂质过氧化(LP)发生条件及反应历程进行了探讨。结果表明:在327℃水浴,微粒体蛋白浓度为1.0mg/ml,pH值7.4的0.1mol/LTris-HCL缓冲液的条件下,150μmol/LNADPH和4.45μmol/l/LFeSO4可使微粒全中丙二醛(MDA)含量于反应15分钟内迅速上升,30分钟时达到高峰,以后趋于平稳。揭示一定比例的NADPH/FeSO4可有效地激发微粒体LP。The condition and the time coures of lipid peroxidation(LP)induced by NADPH/FFeSO 4 in rat hepatic microsomes in vitro were studied.The result revealed that malonaldehyde(MDA)content in microsomes rapidly increased within 15 minutes after addition of 150 μmol/L NADPH and 4.45μmol/L FeSO 4,reached maximal level at 30 minutes,and became constant over the next one hour,when microsomes were incubated at 37℃ in 0.1μmol/L Tis-HCI buffer C=1.0mgPr/ml.pH=7.47.This result suggested that appropiate scale of NADPH/FeSO 4 could effectively induce microsomal liopid peroxidation.A model of enzymatic lipid peroxidation was preliminarily founded in our experiment.
