摘 要:目的了解北京、石家庄、温州和深圳4城市慢性乙型肝炎(乙肝)病毒(HBV)感染者中B基因亚型(Ba和Bj)的分布情况。方法应用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP),对北京(18例)、石家庄(22例)、温州(34例)、深圳(27例)市101例乙肝患者血清标本,经多对型特异性引物巢式PCR鉴定为B基因型慢性HBV感染者的血清标本进行B基因亚型分析,并对部分血清标本以PCR产物直接测序法确定其亚型,以验证PCR-RFLP的准确性。结果经PCR-RFLP分析101例均为Ba亚型,未发现Bj亚型,其中30例经测序分析也证实为Ba亚型。结论4城市的HBV/B主要为Ba亚型。Objective To investigate the distribution of subLgenotype B on hepatitis B virus (HBV) in patients with HBV chronic infection from 4 cities (Beijing, Shijiazhuang, Wenzhou and Shenzhen) of China. Methods A typeLspeeifie nested polymerase chain reaction(PCR) with multiplex pairs of primers was used for HBV genotyping,and Ba and Bj sub-genotypes were identified by a PCR-restrietion fragment length polymorphism (PCR-RFLP) method. A total of 101 serum samples collected from patients with chronic HBV/B infection were detected. Among them, 18 were collected from Beijing, 22 from Shijiazhuang,34 from Wenzhou and 27 from Shenzhen. Thirty from the 101 serum samples were randomly selected and analyzed by PCR product sequencing. Results All of the 101 serum samples were identified as sub-genotype Ba,and none of them was sub-genotype Bj. Conclusion SubLgenotype Ba was a predominant strain of HBV/B in patients with chronic HBV infection from these 4 cities.
