摘  要:目的为骨髓间充质干细胞(BMSCs)在体示踪实验提供一种新型、方便、快捷、可靠的标记方法。方法体外实验部分,Fe2O3分析纯饱和溶液与新鲜胎牛血清按一定比例充分混匀,再与豚鼠BMSCs孵育8h即可标记;体内实验部分,将胎牛血清-Fe2O3标记的豚鼠BMSCs注射到SD大鼠胸腺内,并将大鼠胸腺切片染色观察。结果胎牛血清-Fe2O3标记BMSCs的标记率达到100%,主要标记部位在细胞浆;胎牛血清-Fe2O3标记的BMSCs仍然具有向胸腺被膜细胞、胸腺细胞及血管壁细胞等细胞分化的功能,即对细胞的生存、生长、分化等生物学特性没有影响。结论胎牛血清-Fe2O3可以标记BMSCs,且对BMSCs的生物学特性没有影响。Objective To provide a novel credible technique to label Bone Mesenchymal Stem Cells (BMSCs) directly and conveniently in vivo tracer Study. naethods Ex-vivo experiment: The analytical reagent of Fe2O3 saturated solution intermix with foetal bovine serum for two hours, and mixed with DMED medium in which BMSCs of cavia were incubated for 8 hours. In-vivo experiment: These Being labelled BMSCs were inoculated into thymuses gland of SD rats and being sliced and dyed with perl solution seven days later. Results Dyed by Prussian Blue and Neutral red, these BMSCs can be marked by foetal bovine serum-Fe2O3, and the ratio is about 100 %. The labelled position is in cytoplasm. The marked BMSCs also keep the function of the differentiation into membrane cell, thymus cell and vascular wall cell in the thymus of rats. Also the viability and proliferation of the being marked BMSCs remained unaltered. That is to say, there is no adverse effect on the biological characteristics. Conclusion The BMSCs can be labeled by foetal bovine serum-Fe2O3, and their biological characteristics remains changed.
