摘 要:目的通过HPLC法考察不同外植体对虎杖愈伤组织中大黄素含量的影响。方法选取MS+6-BA0.2mg/L+KT0.2mg/L+2,4-D0.5mg/L为培养基,分别以虎杖的叶片、茎段、叶柄为外植体,诱导出愈伤组织;采用Diamonsil-C18(250mm×4.6mm,5μm)色谱柱;流动相:甲醇-0.1%磷酸(80∶20);检测波长:254nm;流速:1.0mL/min;柱温:27℃;进样量:20μL。结果由叶片、茎段、叶柄所诱导的愈伤组织中大黄素含量分别为0.7096、1.6800、0.1668mg/g,大黄素在0.25~5μg/mL浓度范围内线性关系良好,r=0.99985,平均加样回收率为101.19%,RSD=1.20%。结论HPLC法可用于愈伤组织中大黄素的含量测定,且由茎段所诱导的愈伤组织中大黄素含量较高。Objective To investigate the effect of different explants on emodin contents in the callus of Polygouum cuspidatum by reverse-phase HPLC. Methods The leafs, stems and leaf stalks of Polygouum cuspidatum were used as explants and induce the callus by MS medium containing 6-BA 0.2 mg/L, KT 0.2 mg/Land 2,4-D 0.5 mg/L. The contents of emodin were analyzed by HPLC. The separation was performed on Diarnonsil-Cl8 (250 mm× 4.6 mm, 5 μm) column, using methanol-0.1% H3PO4 (80 : 20) as mobile phase, detected at 254 nm. The flow rate was at 1.0 mL/min, the column temperature was 27 ℃. Results The contents of emodin were 0.709 6, 1.6800 and 0.1668 mg/g in the callus induced by leafs, stems and leaf stalks. The linear range of emodin was 0.25-5 μg/mL, r=0.999 85, the average recovery was 101.19% and RSD was 1.20%. Conclusion HPLC method could be used for the contents determination of emodin in Polygouum cuspidatum callus and the contents of emodin was high in the callus induced by stems.
