摘  要:目的研究3种前体化合物和几种非生物诱导子对丹参(Salvia miltiorrhizaBge)不定根生长及其有效成分含量丹参酮ⅡA和原儿茶醛的影响。方法通过添加前体化合物和诱导子两阶段培养丹参不定根,探讨其对丹参不定根生长和丹参酮ⅡA、原儿茶醛含量的影响。结果培养第0天饲喂前体化合物,发现添加1.0 mmol.L-1乙酸钠、0.5 mmol.L-1酪氨酸和0.5 mmol.L-1乙酸钠分别得到了最高的丹参不定根增殖倍数、丹参酮ⅡA含量和原儿茶醛含量。培养第16天各种非生物诱导子处理,发现超声处理丹参不定根产量和原儿茶醛含量明显增加;1.0 mg.L-1水杨酸促进丹参酮ⅡA合成;高盐和高糖促进丹参不定根生长和次生代谢物合成;硝酸银和硝酸镧促进丹参酮ⅡA合成,但抑制了原儿茶醛合成。结论前体物和诱导子两阶段培养对于丹参酮ⅡA和原儿茶醛合成有显著影响。OBJECTIVE To investigate the effects of precursors and elicitors feedings on the growth of Salvia miltiorrhiza adventitious roots and the contents of tanshinone ⅡA and protocatechuic aldehyde.METHODS The adventitious roots were obtained through tissue culture by feeding precursors and elicitors,and the contents of tanshinone ⅡA and protocatechuic aldehyde were determined by HPLC.RESULTS The highest root growth rate and the content of tanshinone ⅡA and protocatechuic aldehyde were acquired by feeding precursors of 1.0 mmol·L^-1sodium acetate,0.5 mmol·L^-1 tyrosine and 0.5 mmol·L^-1 sodium acetate on day 0 during cultivation.Some kinds of abiotic elicitors were fed on day 16 during cultivation.The results showed that ultrasonic stimulation advanced root growth and protocatechuic aldehyde accumulation,1.0 mg·L^-1 salicylic acid treatment was favorable for tanshinone ⅡA synthesis,the addition of high sugar and high salt greatly improved root growth and secondary metabolite yields,and the addition of silver nitrate and lanthanum nitrate advanced tanshinone ⅡA formation while inhibited protocatechuic aldehyde formation.CONCLUSION Tissue culture by feeding precursors and elicitors had distinct effects on the biosynthesis of tanshinone ⅡA and protocatechuic aldehyde.
