摘 要:目的:研究阿魏酸钠对大鼠缺血性结肠炎(IC)的治疗作用,并探讨其可能的作用机制。方法:用光化学法建立大鼠IC模型,观察不同剂量的阿魏酸钠(20,40,80 mg.kg-1,iv,给药5 d)治疗后大鼠结肠黏膜损伤指数(LI)、内皮素(ET-1)含量、过氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的变化,分析肿瘤坏死因子(TNF-α)、细胞间黏附分子(ICAM-1)在结肠组织的表达情况。结果:与生理氯化钠溶液阴性对照组比较,阿魏酸钠40和80mg.kg-1组LI均显著降低,SOD活性显著升高,而MDA和ET-1含量及TNF-α和ICAM-1的阳性细胞数均显著降低。结论:阿魏酸钠对IC有较好的治疗效果,可以降低结肠损伤。其作用机制可能为拮抗ET-1的生物学效应以及减轻炎症反应与氧自由基的产生。Objective:To determine the effect of sodium ferulate (SF) on ischemic colitis (IC) in rats and its possible mechanism. Methods: IC was induced by photochemistry methods in rats. The effect of SF (20, 40 and 80 mg· kg^-1 ) was intravenously injected for 5 continuous days. Then, changes in lesion index ( LI), endothelin-1 ( ET-1 ) , superoxide dismutase (SOD) , malonyldialdehyde (MDA) ,tumor necrosis factor-a (TNF-α) and intercellular adhesion molecular-1 ( ICAM-1 ) were determined. Results: In comparison with IC control rats, the SOD activity significantly increased; while LI, ET-1,MDA, and the expression of TNF-α and ICAM-1 significantly decreased in SF (40 and 80 mg·kg^-1) -treated rats. Conclusion: SF ameliorates the IC injury in rats, which may be associated with inhibition of ET-1 response, inflammation and the generation of oxygen radicals.
