高效液相色谱法测定饲料和宠物食品中三聚氰胺含量

摘　　要：目的：建立高效液相色谱法测定饲料和宠物食品中三聚氰胺含量的方法。方法：样品经盐酸溶液提取，离心后取5ml上清液于预平衡的Waters Oasis MCX固相萃取柱净化，进行HPLC分析。结果：校准曲线在0．05～1．0mg／L的范围内，相关系数r〉0．999。饲料和宠物食品中三聚氰胺的定量检测限（LOQ）是1．0mg／kg；添加水平为1．0mg／kg时，样品加标回收率为81％～105％，RSD〈10％。结论：方法简便可行，净化效果好，检测灵敏度、准确度及精密度均满足检测要求。Objective：To establish a SPE-HPLC method for the determination of Melamine residues in feedstuff and pet food products. Methods：The Melamine in sample was extracted with Perchloric acid. After centrifugation, the 5 ml supernatant liquid was loaded on an preconditioned Waters Oasis MCX cartridge, and detected with HPLC method. Results-The calibration curve was linear in the range 0.05-1.0 mg/kg and r〉0.999. The limit of quantification （LOQ） was 1.0 mg/kg for Melamine residues in feedstuff and pet food, and at the corresponding level of 1.0 mg/L, The recovery rates were 81% - 105% and RSD 〈 10%. Conclusion ：The method is applicable for the detection of Melamine in feedstuff and pet food. The clean-up effect, sensitivity, accuracy and prevision have all met the analysis requirement.
