摘 要:目的:建立精液标本采集、菌落计数、分类培养、鉴定和杂菌污染判断等相对较为完整的精液细菌培养方法,了解供精者精液中细菌菌落数及其分布情况。方法:对61例精液进行细菌菌落计数,75例精液进行需氧菌及厌氧菌的分类培养。结果:7例精液被杂菌污染,占11.2%;在未被杂菌污染的精液中,90.7%的精液有细菌生长,其中7.4%的精液菌落数>104CFU/ml。细菌分类培养发现精液中以棒状杆菌、草绿色链球菌、表皮葡萄球菌、肠球菌、溶血葡萄球菌、微球菌为主,分别为68.0%、29.3%、20.0%、10.7%、9.3%、8.0%;另外,还培养出缓症链球菌、大肠埃希氏菌、消化链球菌、肺炎克伯雷氏菌、松鼠葡萄球菌、铜绿假单胞菌等精液中稀有细菌或致病菌。结论:精液中细菌常规培养的阳性率高,细菌种类多。Objective: To establish a conventional semen bacterial culture protocol including specimen collection, bacterial count, culture, identification and determination of bacterial contamination, investigate bacterial count and bacterial distribution in the semen of donors, and provide evidence for means to cure male infertility. Methods: Bacterial count was performed in semen specimens from 61 donors, and semen specimens from 75 donors were cultured with and without oxygen. Results: Bacterial culture of the semen was positive in 50(90.7%) of the 54 donors, in whom 5(7.4%) donors had semen bacteria counts 〉 10,000 colony forming units (CFU)/mL except 7 (11.2%) donors whose semen were contaminated. Common bacteria isolated from semen were Corynebacterium(68.0%), α-Hemolytic streptococcus(29.3%), Staphylococcus epidermidis(20.0%), Enterococcus(10.7%), Staphylococcus haemolyticus(9.3%) and Micrococcus Cohn (8.0%). Pathogens and bacteria not often detected in the semen were also discovered, such as Streptococcus mitis, Escherichia coli, Peptostreptococcus, Klebsiella pneumoniae, Staphylococcus sciuri and Pseudomonas aeruginosa. Conclusion: Many different bacterial strains can be detected in semen of donors by culture.
