摘 要:以温州盘菜子叶-子叶柄为外植体,研究不同激素配比、AgNO3浓度、琼脂浓度和苗龄对不定芽再生的影响。结果表明:0.8-1.0 mg/L NAA效果较好,2,4-D浓度大于0.2 mg/L就会抑制不定芽再生,AgNO3浓度以6.0 mg/L最佳,苗龄以5 d为宜,提高琼脂浓度对盘菜不定芽再生有抑制作用。最佳激素配比6-BA 3.0 mg/L+NAA 1.0 mg/L+AgNO36.0 mg/L,不定芽再生率可达68.3%。Cotyledons with petiole from Brassica campestris L. ssp. Rapifera. Matzg are used as explants in vitro culture. Many factors indicate that influence shoots regeneration from cotyledon with petiole, such as hormones combinations, AgNO3 concentration, seedling stage as well as agar concentration . The results show that shoots regeneration can be induced effectively by 0.8-1.0 mg/L of NAA and inhibited by 2,4-D at a concentration higher than 0.2 mg/L. 5-day-old seedling is better, 6.0 mg/L of AgNO2 can induce more adventitious shoots regeneration. High concentration of agar can inhibit adventitious shoots regeneration. Maximum shoots regeneration is obtained in MS medium complementing with 3.0 mg/L 6-BA, 1.0 mg/L NAA and 6.0 mg/L AgNO3, and the regeneration rate reaches 68.3%.
