辅助性T细胞亚群在肝移植患者术后早期变化的研究

摘　　要：目的分析肝移植患者术后早期外周血中Th1/Th2细胞亚群的变化及其在免疫反应中的作用。方法采用细胞内细胞因子免疫荧光染色技术和流式细胞仪检测技术,在单细胞水平上检测11例未发生排斥反应的肝移植患者外周血中Th1/Th2的动态变化。结果11例患者外周血中Th1/Th2在术后3d、1周及2周分别为3.16±2.06、1.83±1.49和2.13±1.80,与术前(5.04±5.27)相比无显著性差异(P﹥0.05),术后3周时Th1/Th2为1.49±1.15,与术前相比显著下降(P﹤0.05)。结论Th1/Th2的平衡与肝移植术后免疫反应密切相关,当Th1/Th2向Th2偏移,易发生免疫耐受反应。Objective To analyze the change of Th1/Th2 cell subset in peripheral blood from patients with liver transplantation. Methods The changes of Thl and Th2 of peripheral blood of 11 patients with liver transplantation were detected on the single cell level by cytokine immune fluorescence staining and flow cytometry. Results There was no significant difference of Th1/Th2 in the first two weeks after operation in the non-AR group. But we found that the Th2 was elevated and Th1/Th2 significantly declined in the third week. Conclusions The ration of Th1/Th2 is correlated with the acute rejection and immune tolerance. Recipients in whom Th2 is predominant are significantly prone to develop immune tolerance.
