摘  要:目的研究酮色林(Ketanserin,KT)对活化的淋巴细胞细胞因子分泌的调节。方法用双抗体夹心ELASA法检测KT对脂多糖(LPS)激活的体外培养的人外周血淋巴细胞细胞因子IL-2、TNF-α、IL-10和TGF-β1分泌的影响。结果KT浓度依赖性抑制LPS激活的人淋巴细胞致炎因子IL-2的分泌,100μmol/L的KT也可抑制致炎因子TNF-α分泌,而10和100μmol/LKT可分别浓度依赖性的提高LPS激活的淋巴细胞分泌抗炎因子IL-10的产量,只有100μmol/L KT可显著提高LPS激活的淋巴细胞分泌抗炎因子TGF-β1。结论KT可对体外活化的淋巴细胞细胞因子分泌进行受体非依赖性的直接调节。Objective To investigate the modulating effects of Ketanserin (KT) on cytokine production of activated lymphocyte in vitro. Methods The levels of IL-2, TNF-α, lL-10, and TGF-β1 produced by in vitro cultured and lipopolysaccharide-activated hmnan peripheral blood lymphocytes were detected with ELISA. Results The 10 and 100 μmol/L KT could decrease the production of IL-2 in lipopolysac- charide-stimulated hmnan peripheral blood lymphocytes. The 100μmol/L KT could also decrease the production of TNF-α, while 10 and 100μmol/L KT could further increased the production of IL-10 in lipopolysaccharide-sfimulated human peripheral blood lymphocytes, and only 100μmol/L KT could further increased the production of TGF-β1. Conclusion KT could modulate directly the production of cytokine of in vitro activated lymphocyte in a receptor-independent manner.
