摘  要:目的探讨促血管生成素-2(Ang-2)对人宫颈癌Hela细胞裸鼠种植瘤血管形成的影响。方法2004年1月至2006年12月在哈尔滨医科大学将实验动物裸鼠分4组:Hank平衡盐溶液组(n=8)、Hela空白对照组(n=9)、Ang-2重组质粒组(n=9)、pcDNA3载体质粒对照组(n=9),比较各组裸鼠成瘤体积、微血管密度(MVD)。结果Ang-2重组质粒组肿瘤体积为(296.72±84.56)mm3,明显小于Hela空白对照组及pcDNA3载体质粒对照组,差异具有统计学意义;Ang-2重组质粒组肿瘤组织中MVD约为12/HP,明显少于Hela空白对照组及pcDNA3载体质粒对照组,差异具有统计学意义。结论Ang-2转染人宫颈癌Hela细胞在裸鼠体内的致瘤力明显降低,肿瘤的体积、肿瘤组织MVD显著低于对照组,表明Ang-2可通过抑制肿瘤血管生成而显著抑制肿瘤生长。Objective To study the effects of angiopoietin-2,which was transfected into the human cervical carcinoma cell line Hela,on the implant carcinoma angiogenesis of nude mouse. Methods From Jan. 2004 to Dec. 2006, the animals were divided into four groups at the First Clinical College of Harbin Medical University: Hank's Balanced Salt Solution control group, Hela control group,angiopoietin-2 recombinant plasmid group, pcDNA3 plasmid vector control group. The carcinoma volume and microvascular density of each group were compared. Results The tumor size of angiopoietin-2 recombinant plasmid group, which was 296.72 84. 56 mm^3 in average, was signifieante smaller than the tumor sizes of the other two groups. The average microvascular density score of the tumors in angiopoietin-2 recombinant plasmid group was 12 per HP, which was also significantly less than the average scores of control groups. Conclusion Nude mouse experiments show that the tumorigenic capacity of cells transfected has been reduced, and that the carcinoma volume and microvascular density are significantly lower than those of the control group. We conclude that angiopoietin-2 inhibits the growth of carcinoma by its inhibition of carcinoma angiogenesis.
