胰岛素治疗大鼠脑外伤后NF-KB表达的实验研究

摘  要:目的分析胰岛素对大鼠脑外伤后核因子-KB(NF-KB)表达的影响。方法96只大鼠随机分为假手术组、外伤组、外伤后胰岛素干预组-(剂量为2.5U/kg)、外伤后胰岛素干预组二(剂量为2.5U/kg),每组24只。测定每组大鼠血糖、胰岛素含量及NF-KB表达,观察不同剂量胰岛素对脑外伤后大鼠NF_KB表达的影响。结果外伤组及胰岛素干预组一、二大鼠伤后6h即可见NF-KB在脑内表达上调,24h表达达到高峰,72h有所回落,7d进一步下降。血糖、胰岛素含量变化也成相同趋势。同时期外伤组NF-KB表达最高,其余依次为胰岛素干预组一、干预组二。结论胰岛素发挥脑保护作用可能是通过抑制NF—KB的表达而实现的,且保护作用与胰岛素的剂量密切相关。Objective To analyze the impact of insulin on the expression of NF-KB in rats after traumatic brain injury (TBI). Methods Sprague-Dawley (SD) rats (96) were randomly allocated into four groups (n=24 for each group): sham operated group; surgical trauma group;insulin intervention group one (2.5 U/kg); insulin intervention group two (5.0 U/kg). We observed the change of blood glucose,insulin,NF-KB and the changes in the expression of NF-KB in different doses. Results The expression of NF-KB rose after 6 hours, and reached its peak after 24 hours, after this period, the expression of NF-KB would be fall-off. The blood serum insulin and glucose have the same change tendency. At the same time, the expression of NF-KB in surgical trauma group reached its peak, for the rest it was insulin intervention group one and insulin intervention group two in order. Conclusion Insulin may exert its neuroprotective effect through inhibiting the expression of NF-KB and the protective effect is concerned with the dose of insulin.
