黄芪失笑散对实验大鼠梗死心肌内VEGF及bFGF mRNA基因表达的影响

摘　　要：目的:黄芪失笑散对梗死心肌内血管内皮细胞生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)mRNA基因表达影响。方法:取SD大鼠76只,随机分成假手术组、模型组、消心痛组、黄芪失笑散小剂量组、黄芪失笑散大剂量组、消心痛加黄芪失笑散大剂量组、贝复济组。治疗20天后进行冠状动脉结扎造成急性心梗的模型,造模成功后再治疗10天后,处死大鼠,取梗死心肌用RT-PCR法进行VEGF及bFGF mRNA的检测。结果:黄芪失笑散大剂量可有效的使梗死心肌内的VEGF及bFGF的mRNA表达升高,并提示黄芪失笑散疗效呈剂量依赖性。结论:黄芪失笑散对缺血性心肌保护作用源于冠脉侧枝循环血管新生。Objective ：The experiment investigates the mechanism of Astragalus Shixiao decoction for promoting angiogenesis through observing its effect on the expression of VEGF and bFGF mRNA in the cardiac muscle of rats in that coronary arteries had been ligated, and provides an experimental basis for further developing and using it in clinic. Methods ： Seventy six SD rats were randomly divided into seven groups ： sham - operated group, model group, Isosorbide dinitrate group, small- dosage Astragalus Shixiao decoction group, large- dosage Astragalus Shixiao decoction group, Isosorbide dinitrate adding large - dosage Astragalus Shixiao decoction group and bFGF group. After 20 days, the models of acute myocardial infarction were made by ligating the left anterior decending branch of coronary artery in rats. The models were made successfully as soon as ST elevated when ligation. The models＇rats were executed, and VEGF and bFGF in infarctional myocardium of each groups were tested by RT - PCR method after ten days of ligation. Results ： Astragalus Shixiao decoction can increase the expression of VEGF and bFGF mRNA in infarctional cardiac muscle of the rats in that coronary arteries were ligated, and the impact is amplified while the dose is enhanced. Conclusion ： Astragalus Shixiao decoction have the protective effect of myocardial Ischemia by the collateral branch of coronary artery.
