摘  要:目的:观察β-淀粉样蛋白1-40(Aβ1-40)大鼠海马双侧注射后大脑组织超氧化物岐化酶(Superoxide Dismutase,SOD)活力和丙二醛(malondialdehyde,MDA)含量的变化及补肾活血化痰法(加减地黄饮子)干预作用。方法:采用大鼠海马立体定向注射凝聚态Aβ1-40诱导老年性痴呆(Alzheimer’sdisease,AD)动物模型。通过分光光度法检测大鼠大脑组织SOD和MDA的活性。结果:与假手术组比较,模型组脑组织SOD活力明显降低(P<0.05),MDA含量明显升高,而加减地黄饮子组脑组织SOD活力较模型组升高(P<0.05),MDA含量较模型组比较明显降低(P<0.05)。结论:加减地黄饮子能增强清除氧自由基的能力,减轻氧自由基引起的脑组织损伤。Objective -To observe effects of the method for Method of Tonifying Kidney and Activating Blood and Dissipating Phlegm on SOD activity and the content of MDA in AD rats induced by amyloid - β1-40 injection into brain. Methods: AD model in rats was incuced by amyloid - β1-40 sterotaxis injection the bilateral Hippocampus. Chinese herbs for Method of Tonifying Kidney and Activating Blood and Dissipating Phlegm (Modified Decoction of Rehmanniae) was geven by oral administration for 28 days. The activity of SOD and the content of MDA was detected by spectrophotometry. Results : The content of MDA in the model group [ (9.09 ± 0.41 ) nmol/mgprot ] was significantly higher than that in the blank group[ (5.44 ± 0.81 ) nmol/mgprot ] ( P 〈 0.05) , and that from the Modified Decoction of Rehmanniae group [ (6.52 ± 0.83 ) nmol/mgprot ] was significantly lower than in the model group ( P 〈 0.05 ). SOD activity from the model group [ (52.43 ± 3.25 ) U/mgprot ] was significantly lower than that in the sham group [ ( 67.87 ± 2.38 ) U/mgplot ] ( P 〈 0.05 ), and that from the Modified Decoction of Rehmanniae group [ (64.06±4.13 ) U/mgprot ] was significantly higher than in the model group ( P 〈 0.05). There was no significant difference of SOD and MAO between the Modified Decoction of Rehmanniae group and Donepezil group [(65.29 ±4.37) U/mgprot] ( P 〉0.05). Conclusion : Modified Decoction of Rehmanniae protect brain neurons injuried from Aβ1-40 through increasing activity SOD and and the decreasing content of MDA.
