摘  要:目的研究鱼藤酮对大鼠中脑黑质组织的毒性作用。方法将24只SD大鼠随机分为4组,每组6只,即空白对照组、溶剂对照组(DMSO-PEG-300)、鱼藤酮染毒组(1.0和1.5mg/kg)。染毒大鼠每日背部皮下注射鱼藤酮溶液,染毒时间为28d。分离中脑黑质组织,应用光镜、电镜观察黑质组织多巴胺神经元形态、突触结构及髓鞘的改变;并检测中脑黑质组织中超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-Px)活力及丙二醛(MDA)和谷胱苷肽(GSH)含量。结果大鼠自主活动减少,出现类帕金森病症候群特征;中脑黑质组织多巴胺神经元形态的改变主要是突触结构异常,突触囊泡数量增多,髓鞘变性等。与空白对照组比较,染毒组黑质组织MDA含量升高(P<0.01),GSH含量降低(P<0.01),SOD和GSH-Px活力降低(P<0.01),差异均有统计学意义。结论鱼藤酮可损伤多巴胺神经元,诱导大鼠出现类帕金森样症状,氧化应激可能参与了鱼藤酮大鼠中脑黑质组织的毒性作用。Objective To study the toxic effects of rotenone on substantia nigra of rats. Methods Rotenone was given subcutaneously once a day during 28 days. Substantia nigra was dissected. Morphology of dopamine neuron, synapse and myelin sheath were observed. SOD and GSH-Px activity were assayed; MDA and GSH contents were measured. Results Autonomic activities of rats decreased, similar to Parkinson's Symptoms. Morphology of dopamine neuron changed, synapse structures were abnormal, synapse vesicals increased and myelin sheath degenerated. MDA content increased (P〈0.01) and GSH content decreased(P〈0.01). SOD and GSH-Px activity declined (P〈0.01). Conclusion Dopamine neuron is damaged and symptoms asimilar to Parkinson's Diseases of rats are induced by rotentone. Oxidative stress is involved in the toxic effects of rats treated with rotenone.
