摘 要:应用番鸭胚成纤维细胞和鸡胚成纤维细胞(CEF)交替传代选育出适应CEF繁殖的番鸭呼肠孤病毒弱毒疫苗株。该弱毒株已失去对1日龄雏番鸭的致病性,对CEF的TCIDs。稳定在10^-5~10^-5.5;在雏番鸭体内连续盲传5代,未见毒力返强现象;1日龄雏番鸭免疫不同代次弱毒后7d攻击番鸭呼肠孤病毒强毒,保护率均在88%以上,且不同代次弱毒的外源病毒检验结果均为阴性。上述结果表明该弱毒株无外源病毒污染、安全性好、遗传性稳定、免疫原性强,可用于制备番鸭呼肠孤病毒病活疫苗。An attenuated vaccine strain of Muscovy Duck Reovirus (MDRV), which was suitable for propagating in chicken embryo fibroblast (CEF), was obtained by serial passage alternately between Muscovy duck embryo fibroblast (MDEF) and CEF. The attenuated strain was proven to be not pathogenic to one-day old Muscovy ducklings and the TCID50 to CEF stable between 10^-5.0- 10^-5.5 ; no reversion of virulence for five passages continuously after introduction to one-day old Muscovy ducklings; and negative on the exogenous virus test, The protection rate of the attenuated strain was greater than 88%, when the ducks were challenged with a virulent strain of MDRV 7 days after vaccination, The results indicated that attenuated vaccine strain of MDRV could be used as a live vaccine against MDRV, as it was free from exogenous virus contamination, and exhibited acceptable genetic stability, excellent safety and effective immunogenicity.
