摘 要:目的探讨斯氏狸殖吸虫病宿主体内T淋巴细胞亚群在免疫调节和发病中的作用。方法将35只清洁级Sprague-Dawley(SD)雄性成年大鼠随机分成7组,其中6组每只鼠腹腔注射斯氏狸殖吸虫囊蚴25个,分别于感染后1w、3w、5w、7w、11w、15w取大鼠外周全血,采用流式细胞术(FCM)检测CD4+、CD8+T淋巴细胞的百分率。最后一组为正常对照组。结果大鼠感染斯氏狸殖吸虫后其外周血CD4+T淋巴细胞亚群水平无明显变化;CD8+T淋巴细胞亚群水平感染后均高于正常水平,1w、5w、11w、15w与对照组比较有显著差异(P<0.05),其中5w增加最为显著;CD4+/CD8+细胞比值在感染后均低于正常水平,5w降至最低,5w、7w、11w与对照组比较差异显著(P<0.05)。结论感染斯氏狸殖吸虫的大鼠主要以CD8+T淋巴细胞反应为主,使机体呈免疫抑制状态,有利于斯氏狸殖吸虫的移行、生长和发育。Objective To explore the immunological regulation mechanism of rat infected by Pagumogobimus skrjabini. Methods 30 Sprague-Dawley (SD) rats were intra-abdominally injected 25 metacercarias and randomly divided into 6 groups. 5 rats were remained as normal control. Peripheral blood was collected and the percentages of T lymphocytes were determined by flow eytometry. Results The CD4^ + T lymphocytes in experimental rats were no significant alteration in the whole course of infection. The CD8^ + T lymphocytes, was significantly increased on week 1, 5, 11, 15 and reached a peak on week 5 after infection ( P 〈 0.05 ). The ratio of CD4 ^+/CD8^ + cells decreased on week 5, 7, 11 after infection ( P 〈 0. 05). Conclusion CD8^ + cell is the main response in rats of infection. The rats exhibit an immune inhibition response, which is very useful in migration, growth, and development of Pagumogonimus skrjabini.
