摘 要:目的钉螺经血水草生物碱水溶液浸泡后,观察肝脏超微结构的变化,探讨血水草生物碱杀灭钉螺的作用机理。方法用血水草生物碱浸泡钉螺48h,解剖钉螺,取肝脏用常规方法制作电镜切片,用日立H-600型透射电镜观察组织结构变化。结果血水草生物碱浸泡后,钉螺肝细胞肿胀、变性,内质网扩张呈空泡状。线粒体嵴肿胀、模糊不清,部分嵴断裂或丢失,呈空泡变;毛细胆管扩张;部分细胞核坏死出现溶解或萎缩;绒毛减少、不规则或消失。结论肝细胞严重损伤是血水草生物碱致钉螺死亡的重要原因之一。Objective To explore the mechanism of eomecon chionantha hance alkaloids(ECA) against Oncomelania hupensis by means of observing the effects of ECA on the liver cell ultrastructure of O. hupensis. Methods Oncomelania hupensis was dissected after immersing in ECA solution for 48 hrs, liver tissues were dissected to prepare conventional electron microscopy specimens and the changes of liver cell ultrastructure were examined by Hitach H- 600 Transmission Electron Microscope. Results After immersing in ECA solution; the liver cell was swollen and degenerated. The rough endoplasma reticulum (RER) expanded, and some of the mitochondrial ridges were swollen, destroyed, Vesiculated and disappeared. The bile canaliculus lumen was enlarged. Some of the cell nuclei were atrophied, coagulated or dissolved, microvilli between the cells near the gland channel diminished and became irregular and vanished. Conclusions The liver cell ultrastructure of O. hupensis is severely damaged after immersing in ECA solution. The results suggest that damaged liver cell ultrastructure is one of the most important killing mechanisms on O. hupensis.
