摘  要:目的改进目前体外扩增树突状细胞(dendritic cell,DC)的方法,探讨含CpG序列的寡脱氧核糖核苷酸(CpG Oligodeoxynucleotide,CpG-ODN)促进DC成熟的作用。方法用500u/ml重组小鼠粒细胞巨噬细胞集落刺激因子(rmGM-CSF)和200u/ml重组小鼠白细胞介素-4(rmIL-4)诱导小鼠骨髓细胞体外分化成DC,采用相差显微镜、扫描电镜、FACS检测细胞表面分子和混合淋巴细胞反应(MLR)等方法比较CpG-ODN、TNF-α等不同培养方法促进DC成熟的作用。结果培养10d,可获得大量典型的DC,经FACS检测细胞表面分子、MLR等证实,CpG-ODN具有较强的促进DC成熟的作用。结论CpG-ODN可以有效促进DC的功能成熟,可应用于科研及临床中。Objective To improve the method of the amplification of dendritic cells(DCs) and to observe the stimulatory effect of CpG-ODN on the maturation of the murine bone marrow-derived dendritic cells. Methods Mice bone marrow-derived DCs were induced in RPMI-1640 with 10% FCS, 500u/ml rm-GM-CSF and 200u/ml rmIL-4. The stimulatory effect of CpG-ODN on the maturation of DCs was evaluated by morphology, phenotype and alloreactivity of DCs. Results A large number of DCs could be collected after 10d. The stimulatory effect of CpG-ODN on the maturation of DCs was confirmed with detecting the expression of molecules on the surface of DCs by FACS and MLR. Conclusion CpG-ODN can effectively enhance the functional maturation of DCs. This method can be used in research and clinic,
