摘  要:目的观察猫角膜内皮细胞(CEC)与人脉络膜黑色素瘤细胞(OCM-1)共培养后增殖能力及细胞周期的变化。方法将角膜内皮细胞分别与肿瘤细胞OCM-1、兔角膜基质细胞在Transwell体系中共培养,通过流式细胞术观察内皮细胞增殖能力及细胞周期的变化。结果与空白对照组角膜内皮细胞周期比较,肿瘤细胞组与基质细胞组角膜内皮细胞S期细胞比例增加,G1期细胞比例下降,肿瘤组增加幅度高于基质细胞组的。肿瘤细胞组角膜内皮细胞S期细胞比例比空白对照组的平均增加近19%,增殖能力显著提高。结论与肿瘤细胞OCM-1共培养能促进角膜内皮细胞增殖。Objective To study the changes in the proliferation and cell cycle of cat corneal endothelial cells (CECs) co - cultured with choroidal melanoma cells ( OCM - 1 ). Methods The Transwell co - culture systems were divided into there groups: experimental group, positive control group and control group. CECs were co -cultured with OCM - 1 cells in the experimental group. CECs were co - cultured with rabbit corneal stromal cells in the positive group. CECs were cultured alone in the control group. The changes in the proliferation and cell cycle of cat corneal endothelial cells were examined by flow cytometry after co - culture. Results Compared with the control, the portions of CECs in S - phase of OCM - 1 group and stromal - cells group increased and the portion of cat endothelial cells in G1 - phase of the two groups decreased. Changes of cell cycle of CECs in OCM - 1 group were more significant than in stromal - cell group. The portion of CECs in S - phase of OCM - 1 group increased nearly 19%, indicating a significant enhancement of cell proliferation. Conclusion CECs co - cultured with OCM - 1 cells can promote proliferation.
