摘 要:目的:探讨HGVNS3区基因变异与基因型的关系。方法:对32株NS3区序列进行酶切位点分析,比较中国株与GBV-C株和已发表的HGVNS3区酶切位点,选择特异性内切酶切点。结果:32株NS3序列酶切位点分析表明存在5种基因型。1组:在4338位无HhaI切点,而在4360位有HhaI切点;2组:4338位及4360位均无HhaI切点;3组:仅在4338位有HhaI切点;4组:4338位及4351位有HhaI及Tsp509I切点;5组:4351位有Tsp509I切点。其核苷酸变异率分别为0.85%、19.0%、20.5%、18.2%、19.6%等。结论:中国株与西非、东非、加拿大、美国及日本株不是同一基因型。Objectives: To explore the relationship between genomic variation and genotypes .in HGV NS3region. Methods: 32 isoates of partial NS3 sequences were analyzed for endonuclease recognition sites. Aftercomparison of Chinese isolates with reported isolates, specific endonuclease recognition sites were selected.Results: In the analysis of endonuclease recognition sites in 32 isoates of partial NS3 sequences, it was foundthat there existed 5 genotypes. There was Hha I recognition site in 436O, and not in 4338 in genotype 1; Ingenotype 2, there was no Hha I recognition site in both 4338 and 4360; In genotyPe 3, there was Hha Irecognition site in 4338; In genotype 4, there were Hha I recognition site in 4338 and Tsp5O9 I recognitionsite in 4351; In genotype 5, there was Tsp5O9 I recognition site in 4351. In the 5 genotypes, their ntvariation rates were 0. 85 %, 19. O %, 2O. 5 %, 18. 2 %, 19. 6 % respectively. Conclusions: Our studysuggested that isoates from China were not of the same genotype as those from West Africa, East Africa,Canada and America.
