摘 要:目的:观察单抗PF18-3对小鼠胸腺树突状细胞系(MTSC4)增强胸腺细胞凋亡作用的影响并对PF18-3识别分子进行特征分析。方法:用DNA梯度断裂片段电泳检测PF18-3对凋亡过程的抑制作用;用膜蛋白斑点杂交和免疫共沉淀对其识别分子进行特征分析。结果:PF18-3抑制MTSC4对胸腺细胞凋亡的增强作用。其识别分子为分子量31000的膜蛋白,在不同胸腺基质细胞系分布不同,与其凋亡介导增强作用一致。结论:PF18-3识别分子参与凋亡介导增强过程,为非凋之介导分子CD95/配体(Fas/FasL)分子,值得进一步研究。Objective: Observation of effect of PF18-3 monoclonal antibody on the enhancement of thymocyteapoptosis by mouse thymic dendritic cell line (MTSC4) and characterization of PF18-3 recognized molecule.Methods: DNA fragmentation assay was used to observe effect of PF18-3 on the process of apoptosis while dotblot of membrane protein and immunoPrecipitation were conducted for characterization of the PF18-3recognized molecule. Results: PF18-3 depressed the enhancement of thymocyte apoptosis by MTSC4. Theimbalance distribution of PFl8-3 recognized molecule, a 31 000 protein, on the surface membranes of twodifferent mouse thymic stromal cell lines was consist with its apoptosis-inhibiting function. Conclusion:Results in this study indicate the involvement of PF18-3 recognized molecule in the enhancement by MTSC4 ofthymocyte apoptosis. Also PFl8-3 recognized molecule with a molecule weight different from Fas and Fasligand deserves to be further studied.
