摘  要:通过MTT比色法和Westernblot法检测NasSeV5O18·3H2O的抗肿瘤活性及作用机理.结果表明,Na5seV5O18·3H2O(0.625-20mg·L^-1)能抑制K562细胞增殖(P〈0.05);药物处理K562细胞24h,可使胞浆细胞色素C,IκBα含量明显增多,核内NF-κB含量减少.结果提示Na5SeV5O36·3H2O能体外抑制K562细胞增殖,其机理可能与线粒体凋亡和NF-κB/IκBα信号转导有关.Aimed to study the effect and mechanism of selenium compound Na5SeV5O18 · 3H2O, MTT colorimetric method and Western blot were used to test anti-proliferation activity and the content of cytochrome C,NF-κB and IκBα in K562 cells. The results show that Na5SeV5O18 · 3H2O 0. 625~20 mg · L^-1 could significantly inhibit proliferation of K562 cells in vitro in a time and dose-dependent manner, IC50 values were 14.41 and 3.45 mg · L^-1 after treated for 48 h and 72 h, respectively, and the intracellular content of cytochrome C and IeBa markedly increased and NF-κB decreased when treatment for 24 h. The results imply Na5SeV5O18 · 3 H2O has anti-tumor activity and the mechanism of which may relate to apoptosis of mitochondria and signal pathway of NF-κB/ IκBα.
