摘 要:目的建立测定人血浆中泮托拉唑的HPLC法。方法血浆样品经液-液萃取后,以甲醇-水(体积比为60:40)为流动相,倍他米松为内标,采用Diamonsil C18柱(4.6mm×150mm,5μm)分离,在288nm处进行检测。结果标准曲线线性范围为0.02~5.0mg·L^-1,定量下限为20μg·L^-1,日内和日间精密度(RSD)均小于8.4%。准确度在104.3%以内,低、中、高3个质量浓度的提取回收率分别为75.4%、64.8%和65.3%。结论该法操作简便、快速、灵敏,适用于泮托拉唑的临床药物动力学及制剂的生物等效性评价。Objective To develop an HPLC method for the determination of pantoprazole in human plasma. Methods After a simple liquid-liquid extraction, pantoprazole and the internal standard were separated on Diamonsil C18 column and detected at the wavelength of 288 nm. The mobile phase consisted of methanol- water ( V: V = 60 : 40). Results The linear calibration curve was obtained in the concentration range of 0.02 - 5.0 mg·L^-1. The lower limit of quantification was 20 μg·L^-1. The intra-and inter-day precision (RSD) were less than 8.4 %, and the accuracy was within 104.3 % calculated from QC samples. The extraction recoveries of pantoprazole at different levels (low, medium, high) were 75. 4%, 64.8% and 65.3 %, respectively. Conclusions The method is proved to be suitable for the clinical investigation of pharmacokinetics of pantoprazole, which offers the advantage of simplicity, speed and sensitivity.
