摘 要:目的探寻幽门螺杆菌(Hp)促细胞增殖组分。方法利用超声破碎法提取NCTC11637Hp不同组分,获得分泌成分、菌膜及胞浆内容,分别加入SGC-7901细胞,共育24h,用MTT法检测各组分对细胞增殖的影响。进而用细菌蛋白提取试剂盒(P-PEK)分离菌膜蛋白,获得4个不同溶解度膜蛋白组分,分别或两两组合后加入SGC-7901细胞,用MTT法和Ki-67细胞免疫组化检测细胞增殖情况。结果低浓度Hp菌膜(≤1μg/mL)可促细胞增殖,增殖率可达119.32%(P<0.05);Hp菌膜3个不同溶解度膜蛋白组分单独作用于细胞时抑制细胞增殖,但低浓度的"中溶解度"和"难溶解度"混合组分促细胞增殖,增殖率可达115.55%(P<0.05)。结论Hp促细胞增殖成分存在于菌膜的"中溶解度"和"难溶解度"混合组分中。The effect of the membranous component isolated from Helicobacter pylori to promote the proliferation of human gastric adenocarcinoma SGC-7901 cells was investigated in the present study,in which the secretory,membranous and cytoplasmic components from the cag^+ H pylori were obtained by treatment with ultrasonicator and the amount of proteins thus obtained in the components was measured by Bradford-based assay. Meanwhile,the lipopolysaccharide(LPS)was isolated with hot carbolisata aqua and detected by MTT assay. These various components at different protein concentrations or LPS at different doses were co-cultivated with the SGC-7901 cells for 24 hours,and the influence of various components on the cell proliferation was determined by MTT and the immunohistochemical assay with Ki-67 monoclonal antibody. The experimental results showed that all the secretory and cytoplasmic components as well as the LPS of H. pylori could inhibit the proliferation of SGC-7901 cells,in which the higher doses(≥2 5 μg)of cag^+ H pylori membranous protein inhibited cell proliferation,but the lower doses(≤1μg/ml)of the membranous component promoted cell proliferation with a 119.32% growth rate of cells(P〈 0.05) In addition,the cag H pylori membranous proteins could also promote cell proliferation,but its intensity was weaker than that of the cag^+ H pylori one. The membranous components of H pylori with 3 different solubilities were found to be inhibitory to cell proliferation when they were used separately, while the low concentration of mixed membranous protein( partially soluble and insoluble proteins)promoted cell proliferation with a 115.55% growth rate. (P〈0.05). It is evident that the promoting effect of the membranous proteins from H pylori exists in the low concentration of mixed portions, i e the partially soluble and insoluble proteins.
