摘  要:目的 研究阿魏酸钠脑室内注射对谷氨酸单钠诱导的成年小鼠神经损伤的可能的修复作用。方法小鼠接受谷氨酸单钠(MSG,4.0g·kg^-1·d^-1)灌胃,每日1次,连续给药10d,对照组同时接受同体积的生理盐水灌胃。MSG+SF组在MSG灌胃后第1,10,20天侧脑室注射不同剂量的阿魏酸钠(SF)3次,末次SF注射后第11天,开始爬绳和Y迷宫实验;行为学实验结束后,进行脑海马组织病理学检查。结果末次给SF后第30天MSG组会爬绳小鼠数(3/10),明显少于对照组(10/10)(P〈0.01);末次给SF后第26天MSG组Y迷宫正确反应次数(14.6/20),明显少于对照组(17.9/20)(P〈0.01)。MSG组小鼠海马细胞水肿、神经元变性坏死和组织增生。然而,SF侧脑室注射则能部分修复MSG诱导的小鼠行为障碍和组织病理学损伤,且与剂量相关:末次给SF后第30天MSG+SF(10.0me/kg)组会爬绳小鼠数(8/10)较MSG组多(P〈0.05),接近对照组,但在MSG+SF(2.5,5.0me/kg)组效果不显著;末次给SF后第26天MSG+SF(5.0,10.0me/kg)组Y迷宫正确反应次数分别为(16.2/20,16.5/20)明显多于MSG组(P〈0.05),但在MSG+SF(2.5me/kg)组效果不显著;MSG+SF(2.5me/kg)组海马区出现轻微的神经元损伤和组织增生,而MSG+SF(5.0,10.0me/kg)组海马却未发现明显的组织病理学改变。结论阿魏酸钠脑室内注射对谷氨酸单钠诱导的成年小鼠兴奋性毒性神经损伤有一定的修复作用。Objective To investigate a possible brain repair following glutamate-induced neuronal damage mediated by intracerebroventricularly administered sodium ferulate (SF) in adult mice. Methods Mice received intragastric (ig) administration of monosodium glutamate (MSG) (4.0g · kg^-1 · d ^-1 ) for 10 d, and the mice in control group received ig administration of normal saline. The animals in the MSG + SF groups received intracerebroventricular (icy) administration of SF (2.5, 5.0, 10.0mg · kg^-1·d^-1) 1 d, 10 d and 20 d later, and the mice in control and MSG-treated groups received icy injection of DMEM simultaneously. On day 11 after the administration of SF the behavioural tests were performed, and then the histopathology of the animal brains was studied to analyze MSG-induced functional and morphological changes. Results Behavioural tests showed that on day 30 after administration of SF, the number of MSG-treated mice which could crawl along a rope(3/10) was obviously less than that of control(10/10) ( P 〈 0.01 ) ; on day 26 after administration of SF, correct responses of MSG-treated group in Y-maze test ( 14.6/20)were significantly less than that of control(17.9/20) ( P〈0.01 ). Examination of histopathology displayed MSG-induced hippocampal lesions were characterized by intracellular edema, degeneration and necrosis of neurons, and hyperplasia. However, icy administration of SF obviously facilitated brain recovery from glutamate-induced behavioural disturbances and histopathological damage in adult mice in a dose-dependent manner: on day 30 after administration of SF, the number of MSG + SF(10.0mg/kg) -treated mice which could crawl along a rope(8/10) was obviously more than that of MSG ( P〈 0.05 ), close to that of control, but MSG + SF(2.5,5.0mg/kg) did not significantly effect; on day 26 after administration of SF, correct responses of MSG + SF(5.0,10.0mg/kg) -treated mice in Y-maze test (16.2/20,16.5/20) were significan
