摘 要:目的探讨复方中药三甲益肝冲剂(Sanjia-yigan granule,S-JG)对肝纤维化的预防机制和效果。方法雄性SD大鼠40只,随机分为模型组、复方丹参组、S-JG组和正常对照组各10只。模型组、复方丹参组、S-JG组均以二甲基亚硝胺(DMN)腹腔注射诱导大鼠肝纤维化模型;模型组造模同时给予生理盐水灌胃,复方丹参组造模同时给予复方丹参口服液灌胃,S-JG组造模同时给予S-JG灌胃;正常对照组腹腔注射生理盐水并用自来水灌胃,共4周。4周后取肝组织标本,行HE和Masson染色,免疫组织化学法检测肝组织结缔组织生长因子(CTGF)。结果(1)肝脏病理学改变:模型组可见较粗的纤维间隔;复方丹参组、S-JG组可见稍细的纤维间隔。(2)Masson染色胶原纤维面积比较:模型组胶原面积较正常对照组显著增加(P<0.05),复方丹参组和S-JG组胶原面积均较模型组减轻(P<0.05);(3)免疫组织化学检测结果:模型组CTGF强阳性表达,S-JG组和复方丹参组组表达少于模型组。结论三甲益肝冲剂抗纤维化作用与其抑制肝组织中CTGF的表达有关。Objective To explore the mechanisms and preventive effect of Sanjia-yigan granule (S-JG) on hepatic fibrosis. Methods Forty male SD rats were randomly divided into four groups : model group, salvia miltiorrhiza group, S-JG group and control group. The dimethylnitrosame ( DMN ,2 mL · kg^ -1 ,once daily ,3 days serially a week) was injected intraperitoneally for 4 weeks in model group, salvia miltiorrhiza group( SM group) and S-JG group to induce the models of hepatic fibrosis;The S-JG in S-JG group and salvia miltiorrbiza in SM group were respectively given by intragastric administration, and the hepatic fibrosis were induced as model group ;normal saline was given for 4 weeks in control group. The expressions of connective tissue growth factor (CTGF) were observed by immunohistochemical staining. Results ( 1 ) Pathological changes of liver:The fibrosis interval of hepatic tissues in model group was wider than that in SM group and S-JG group, and that in Sm group was equal to that of S- JG group. (2)Comparison of collagen area in Masson staining:The comparison of collagen area in model group were more than that of control group (P 〈 0. 05 ) , The comparison of collagen area in SM group and S-JG group were less than that of model group(P 〈0.05). (3) Results of immunohistochemical staining:The expressions of CTGF were strong positive in model group; The expressions in SM group and S-JG group were lower than that of model group. Conclusion The mechanism of S-JG and salvia mihiorrhiza in anti-fibrosis effects may be correlated with its inhibiting the expression of CTGF in liver tissues.
