摘  要:目的探讨长期游泳运动后大鼠大脑额叶运动相关脑区c-fos的表达。方法雄性Sprague-Dawley(SD)大鼠55只,分为对照组(A组,n=5)、持续运动组(B组,n=25)和间歇运动组(C组,n=25),其中B、C组以一定方式进行游泳运动。训练持续6周,B、C组大鼠在末次运动结束后即刻,30min,60min,120min,240min每一时间点随机选取各5只,A组与即刻组同时经戊巴比妥钠腹腔麻醉,依次灌注生理盐水、4%多聚甲醛固定后取脑。行免疫组化,计数c-fos阳性细胞,进行统计学分析。结果c-fos在初级运动皮质(M1)、次级运动皮质(M2)、伏核(A)呈均匀表达,两种游泳运动后,M1区c-fos表达峰值都出现在30min;间歇组M2区c-fos表达峰值在60min,而持续组M2区表达无显著变化;伏核c-fos表达,在间歇组峰值出现在60min,持续组则呈增高趋势,240min仍未见下降。结论游泳运动后不同时刻大鼠脑运动皮质、伏核c-fos的表达可能与运动疲劳、恢复有关。Objective To explore the effect of long-term swimming exercise on c-fos expression in the regions of frontal lobe related to movement in rat. Methods Fifty-five male Sprague-Dawley rats were divided into control group(group A), endurance exercise group(group B) and interval exercise group(group C). The rats in groups B and C were trained by different types of swimming for 6 weeks. At 0, 30, 60, 120, 240 min after the last swimming exercise, the rats in groups B and C were anesthetized with sodium pentobarbital and perfused transcardially with heparinized physiological saline followed by 4% paraforaldehyde in 0.1 rnmol/L phosphate buffer, while the rats in control group were anesthetized and perfused at the first time point. The brains were removed. The expression of c-fos protein was determined by irnmunocytcchemical method. Results The expression of c-fos in rat primary motor cortex(M1) were maximal at 30 min in groups B and C. In secondary motor cortex(M2) ,the expression of c-fos peaked at 60 min in group C, but there was not obvious changes in group B. In nucleus accumbens(A), the maximum of c-fos presented at 60 rain in group C, however, the expression increased continually until 240 min in group B. Conclusion The expression of c-fos after swimming exercises in rat motor cortex and nucleus accumbens may involve in the mechanism of central fatigue and recovery.
