摘 要:目的研究巨噬细胞株分泌的RCAS1对正常小鼠骨髓单个核细胞(BMMNC)凋亡的影响。方法RT-PCR方法检测巨噬细胞株RCAS1基因表达,10例正常小鼠骨髓基质细胞RCAS1基因表达作为正常对照组。流式细胞术检测8例经巨噬细胞株培养上清作用后正常小鼠BMMNC的凋亡,8例经正常小鼠骨髓基质细胞培养上清作用后正常小鼠BMMNC的凋亡作为正常对照组。对8例巨噬细胞株RCAS1的表达强度与8例经巨噬细胞株培养上清作用后正常小鼠BMMNC的凋亡率进行相关分析。结果巨噬细胞株RCAS1 mRNA的表达强度为(1.98±0.24),明显高于正常对照组(0.84±0.21)(P<0.01)。②巨噬细胞株培养上清作用后正常小鼠BMMNC的凋亡率为(16.66±1.97)%,明显高于正常对照组(2.10±0.25)%(P<0.01)。③巨噬细胞株RCAS1mRNA的表达强度与巨噬细胞株培养上清作用后正常小鼠BMMNC的凋亡率呈正相关(r=0.95,P<0.01)。结论巨噬细胞株高表达RCAS1,RCAS1对正常小鼠BMMNC有促凋亡作用。Objective To investigate the effect of RCAS1 secreted by rnacrophage cell lines on the apoptosis of bone marrow mononuclear cells(BMMNC) in normal mice. Methods Expression of RCAS1 mRNA in rnacrophage cell lines was measured by reverse transcriptase polymerase chain reaction(RT PCR), and 10 cases of bone marrow stromal cells in normal mice were normal controls. The apoptosis of normal BMMNC stimulated by supematant of macrophage cell lines was measured by flow cytometry, and 8 cases of normal BMMNC stimulated by supernatant of bone marrow stromal cells in normal mice were normal controls. The correlation of expression intensity of RCAS1 mRNA in macrophage cell lines and apoptosis of normal BMMNC stimulated by supernatant of macrophage cell lines was analyzed. Results ① Expression intensity of RCAS1 mRNA secreted by rnacrophage cell lines( 1.98 ± 0.24) was higher than that of normal controls(0.84 ± 0.21 ) ( P 〈 0.01 ). ②The apoptosis of normal BMMNC stimulated by supernatant of rnacrophage cell lines( 16.66 ± 1.97) % was al.so higher than that of normal controls (2.10 ± 0.25 ) % (P 〈 0.01 ). ③ Expression intensity of RCAS1 mRNA secreted by macrophage cell lines was positively correlated with the apoptosis of normal BMMNC stimulated by supernatant of macrophage cell lines( r = 0.95, P〈 0.01 ). Conclusion The results indicate that RCAS1 plays a pro-apoptotic role on BMMNC of normal mice.
