摘  要:目的深入探讨半边旗(Pteris semipinnataL,PsL)二萜化合物5F(PsL5F)抗癌作用的分子机制,全面了解其对卵巢癌中癌相关基因表达的影响。方法应用人癌相关基因芯片观察PsL5F对多种癌相关基因表达的影响,并用实时定量PCR和Western blot对部分基因和蛋白进行验证。结果在芯片所含440种基因中,PsL5F明显上调52个基因的表达,明显下调25个基因的表达。差异表达明显的基因可粗略分为3类:影响p53及相关基因;②影响TGF-β通路相关基因;③抑制细胞外基质和粘附分子的表达。结论芯片结果明显深化了对PsL5F抗癌分子机制的认识,其作用机制与p53、TGF-β相关基因及细胞外基质和粘附分子表达有关。Aim To investigate the effect of 5F from Pteris semipinnata L ( PsLSF ) on cancer-associated gene expression in highly metastatic ovarian carcinoma HO-8910PM cells, and explore its molecular mechanism of antitumor effect. Methods The cancer-associted gene expression profiles of HO-8910PM cells with and without PsLSF-treatment were analyzed by using genechip technology. To validate the genechip data, some differential expression genes, including BRCA1, GDF15 and NR1D1 were analyzed with fluorescent quantitative real-time PCR and Western blot assay. Re-suits 77 differently expressed genes were screened out, of which up-regulated and down-regulated genes were 52 and 25, respectively. These genes were related to apoptosis, cell cycle, cell growth and differentiation, cell motility, signal transduction, transcription and metabolism. Conclusions The results indicated that antitumor effect of PsL5F in HO-8910PM cells was associated with p53, TGF-beta signaling pathway, extracellular matrix and adhesion molecule expression.
