摘  要:目的:研究无血清培养条件下SP2/0细胞的相关肿瘤学特性。方法:应用MTT法检测用无血清培养所得的SP2/0细胞的增殖能力及这些细胞中SP细胞的比例,并检测它们的体内致瘤性。结果:SP2/0细胞在无血清培养基中增殖缓慢,但当把它们放回含血清培养中,则表现出比普通培养的SP2/0细胞有更高的增殖能力;无血清培养可以使SP2/0细胞中的SP细胞比例增加;无血清培养的SP2/0细胞比普通培养的SP2/0细胞在鼠体内有更高的体内致瘤性。结论:无血清培养可以增加SP2/0细胞中具有干细胞特性的瘤细胞比例,即可以富集SP2/0细胞的肿瘤干细胞。Objective: To investigate the oncological characters of SP2/0 cells cultivated in serum-free medium. Methods: First, SP2/0 cells were cultivated in serum-free culture solution, then MTT colorimetry method, detecting SP cells, tumorigenesis experiment in BALB/c mice were respectively used to investigate their oncological characters. Results: SP2/ 0 cells proliferated slowly when they were cultivated in serum-free medium. However, when these cells were put back into serum-containing medium, they showed higher reproductive activity than that of SP2/0 cells cultivated in serum-containing medium did. Further, the ratio of SP cells were improved when SP2/0 cells were cultivated in serum-free medium. Last, SP2/0 cells that cultivated in serum-free medium gained higher oncogenicity in vivo. Conclusion: When cultivating in serum-free medium, the ratio of SP2/0 cells possessing the property of stem cells can be increased, what can also be said serum-free culture can enrich tumor stem cells in SP2/0 cells.
