摘 要:目的为筛选出辐照灭菌中评价对病毒灭活效果的指示噬菌体,比较研究了噬菌体T4、ФX174D、MS2和f2,以及两种细菌参考株大肠杆菌8099和枯草杆菌黑色变种芽孢ATCC 9372对电离辐射γ射线的抵抗力。方法采用钻源。60^Co-γ射线辐射源对噬菌体和细菌进行辐照处理;灭活方法采用悬液灭活试验;噬菌体的检测采用双层琼脂法。结果方差分析表明,不同剂量的γ射线组间(P〈0.01)和不同的噬菌体组间(P〈0.05),差异均有统计学意义。LIV(灭活对数值)和KL(杀灭对数值)为:(1)T4、ФX174、f2和MS24种噬菌体经100Gy的γ射线辐照可达到消毒水平(LIV≥4.00log10);在此剂量下4种噬菌体的LIV分别为:6.31,6.92,5.74和4.46log10;(2)在前述剂量下,大肠杆菌8099的KL〉7.97log10;(3)在前述剂量下,枯草杆菌黑色变种芽孢ATCC9372的KL为1.61log10结论6种微生物对γ射线的抵抗力由强到弱为:枯草杆菌黑色变种芽孢〉噬菌体MS2〉噬菌体MS2〉噬菌体f2〉噬菌体T4〉ФX174〉大肠杆菌。Objective To screen the suitable bacteriophage as virus indictor in irradiation sterilization. Methods Suspensions of bacteriophage T4, ФX174D, MS2, and f2, Escherichia coli 8099,and Bacillus subtilis var. niger, sp. ATCC9372 were irradiated with 60^Co-γ ray. The mean log10 inactivation value (LIV)and killing log value(KL) were calculated. Results (1)Under 100 Gy of gamma-radiation, the LIV levels of the bacteriophage T4, ФX174, f2, and MS2 were 6.31,6. 92,5.74, and 4. 46 log10 respectively, all reaching the disinfection level ( LIV ≥ 4. 00 log10 ), (2) Under the same absorbed dose, the KL of Escherichia coil 8099 was 〉 7.97 log10 ; (3) Under the same absorbed dose, the KL of the Bacillus subtilis var. niger, sp. ATCC9372 was 1.61 log10. Conclusion The order of resistance of the above six microorganisms to gamma-radiation from the biggest to the smallest is as follows :Bacillus subtilis var. niger. sp. 〉 bacteriophage MS2 〉 bacteriophage f2 〉 bacteriophage T4 〉 bacteriophage ФX174D 〉 E. coll.
