单次静脉注射咪达唑仑的催眠量效关系

摘　　要：目的研究单次静脉注射咪达唑仑的催眠量效关系。方法将30例ASAⅠ～Ⅱ级全麻病人分为6组,每组5例,分别给予咪达唑仑0.04、0.06、0.08、0.10、0.12和0.15mg.kg-1静脉注射;以睫毛反射消失,大声呼唤无睁眼为入睡标准。采用probit法和logit法计算咪达唑仑的半数有效剂量(ED50)及95%有效剂量(ED95)。结果probit法咪达唑仑的催眠ED50值(95%可信限)为0.088(0.066-0.110)mg.kg-1,ED95值(95%可信限)为0.157(0.120-0.395)mg.kg-1;logit法咪达唑仑的催眠ED50值(95%可信限)为0.088(0.064,0.113)mg.kg-1,ED95值(95%可信限)为0.163(0.121,0.588)mg.kg-1。结论probit法咪达唑仑的催眠ED50值(95%可信限)和ED95值(95%可信限)分别为0.088(0.066-0.110)mg.kg-1和0.157(0.120-0.395)mg.kg-1。Objective To define the median and 95% effective dose of hypnotic of midazolam. Methods Thirty nonpremedical patients, ASA gradeⅠ～Ⅱ, undergoing elective surgery were randomly divided into 6 groups. The patient of each groups received 0.04,0.06,0.08,0.10,0.12 or 0.15mg·kg^-1 of midazolam intravenous bolus respectively. The means values of 50% patients effective dose （ED50） and ED95 were obtained by probit analysis and logit analysis at the endpeint of hypnotic （loss of response to verbal command and loss of eyelash reflex）. Results By probit analysis, the means ED50 and ED95 values of midazolam hypnotic （95% confidence interval） were 0.088 （0.066,0.110） mg·kg^-1 and 0,157 （0.120, 0.395） mg·kg^-1 respectively, By logit analysis, the means ED50 and ED95 values of midazolam hypnotic were 0.088 （0.064, 0.113） and 0,163 （0.121, 0.588）mg·kg^-1 respectively. Conclusions By probit analysis, the means ED50 and ED95 values of midazolam hypnotic （95% confidence interval） were 0.088 （0.066-0.110）mg·kg^-1 and 0.157（0.120-0.395）mg·kg^-1 respectively.
