酵母β-1,3-葡聚糖酶解产物体内抗肿瘤及抗氧化活性的测定

摘　　要：以酵母-β1,3-葡聚糖酶解产物为原料,经超滤得到水溶性葡聚糖WSGI(water soluble-β1,3-glucanI,WSGI)(Mr>10 ku)和WSGII(Mr<10 ku),进行体内抗肿瘤试验,比较不同给药方式以及水溶性葡聚糖与羧甲基葡聚糖对肿瘤小鼠抑瘤率、胸腺指数、脾脏指数的影响,并测定脂质过氧化物丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力等抗氧化指标。结果表明,WSGI能明显抑制肿瘤S180的生长,抑制率可高达49.84%,并且呈现出剂量-效应关系,其抑瘤效果强于同剂量下羧甲基葡聚糖,与环磷酰胺阳性对照组相比差异不显著,腹腔注射给药抑瘤效果好于灌胃给药;同时WSGⅠ可显著增加小鼠体内的抗氧化活性;而分子量较小的WSGII则没有明显的抑瘤效果。Water soluble glucan WSGⅠ[（Mr〉10 ku）and WSGⅡ（Mr〈10 ku）] were separated and purificated by ultrafiltration from yeast glucan hydrolysates. The effect of WSGⅠ in different administration routines,water soluble glucan and carboxylmethyl-glucan were compared on tumor inhibition rate, thymus index,spleen index, and the malondialdehyde （MDA）, superoxide dismutase （SOD） and glutathione peroxidase （GSH-PX） were examined. The results showed that WSGⅠ could remarkably inhibit the growth of S180 in a dosage-dependent manner and the inhibition rate of 200 mg/（kg · d） group was 49.84%. The antitumor activity of WSGⅠ was higher than that of carboxylmethyl-glucan at the same dosage and was close to that of Cy control. The antitumor activity of WSGⅠ by intraperitoneal injection was higher than that by gavage. WSGⅠ reduced the level of MDA and increased the activities of SOD and GSH-Px in liver of mice. WSGⅠ had high antitumor and antioxidant abilities. The antitumor activity of WSPⅡ（Mr〈10 ku）could not be observed.
