中国耶氏肺孢子虫基因型的初步分析

摘　　要：为研究我国耶氏肺孢子虫的基因分型，利用巢式PCR方法扩增24例肺孢子虫肺炎（PCP）患者支气管肺泡灌洗液中的耶氏肺孢子虫（Pneumocystis jiroveci）核糖体DNA内转录间隔区（ITS1-5.8S rDNA-ITS2）基因。纯化回收目的片段，将其克隆到PMD18-T载体，并导入JM109细胞系中，每个标本制备3～5个克隆，提取质粒测序。从19例患者的标本中检出ITS1-5.8SrDNA-ITS2基因，其中12份标本的25个克隆的测序结果可用于基因分型。经与Lee等（1998）的分型标准进行比对，本组病例耶氏肺孢子虫的ITS1分为B、E、I、N4型，最常见的为E型；ITS2分为a、b、e、h、i、n6型，最常见的为i型；ITS基因分9型，以Bi多见，有2份标本存在混合感染。ITS1-5.8S rDNA-ITS2 regions were amplified from BALF specimens from 24 patients with PCP by use of nested PCR. PCR products were purified, cloned to PMD18-T vector, and introduced to JM109 cell line. ITS of 19 specimens were detected. Of them, 25 clones of 12 specimens were available for genotyping. According to the standard of Lee et al. （ 1998）, total of 4 genotypes of ITS1 and 6 genotypes of ITS2 were found. The types of ITS1 were B, E, I, N, and the most prevalent type of ITS1 was E; the types of ITS2 were a, b, e, h, i, n, and the most prevalent type of ITS2 was i. Taken altogether 9 genotypes of ITS were found. The mixed infection was presented from 2 specimens.
