乌骨藤提取物对人结肠腺癌细胞增殖的实验研究

摘　　要：目的:研究乌骨藤提取物对体外培养的人肠癌细胞(Lovo)增殖的影响。方法:用两种方法(A、B)制备乌骨藤提取物,分别采用不同浓度作用Lovo细胞,通过细胞形态学和MTT法检测乌骨藤提取物对Lovo细胞增殖的影响。结果:乌骨藤提取物A对细胞的最大无毒浓度约为0.648 mg/mL;药物浓度在0.389、0.648、1.08、1.8、3 mg/mL时的抑制率分别为6.58%、8.24%、13.02%、59.69%、66.23%,用回归法求出乌骨藤提取物A对Lovo细胞增殖的半数抑制浓度(IC_(50))为1.625 mg/mL。乌骨藤提取物B对细胞的最大无毒浓度约为0.648 mg/mL,药物浓度在0.389、0.648、1.08、1.8、3 mg/mL时的抑制率分别为14.49%、15.48%、29.69%、63.26%、64.05%,用回归法求出乌骨藤提取物B对Lovo细胞增殖的半数,IC_(50)为1.84 mg/mL。结论:乌骨藤提取物对体外培养的Lovo细胞的增殖有明显抑制作用。AIM： To investigate the effects of the extracts from Wu Guteng on the multiplication of Lovo cells （human colon cancer）. METHODS： Two methods（A, B） are used to prepare the medicine solution from the liquids withdrawn of Wu Guteng, which is diluted to different densities to incubate with Lovo cells for 24 h and 48 h. Cytomorphology and the MTT method are used to test the effects of the liquids withdrawn from Wu Guteng on the multiplication of the Lovo cells. RESULTS： The maximal nontoxic density of the extractive from Wu Guteng by method A to Lovo cells was about 0.648 mg/mL. When the medicine density were 0. 389, 0.648, 1.08, and 1.8 and 3 mg/mL, the inhibiting rates were 6.58%, 8.24%, 13.02%, 59.69% and 66.23% respectively. The IC50 of extractive from Wu Guteng by method A to Lovo cells was 1. 625 mg/mL. The maximal nontoxic density of extractive from Wu Guteng by method A ws about 0.648 mg/mL When the density of the extractive from Wu Guteng by method B to Lovo cells were 0.389, 0.648, 1.08, and 1.8 3 mg/mL, the inhibiting rates were 14.49%, 15.48%, 29.69%, 63.26% and 64.05% respectively. The IC50 of extractive from Wu Guteng by method B was 1.84 mg/mL. CONCLUSION： The extracts from Wu Guteng inhibite the multiplication of the cancer cells of colon glands.
