摘 要:目的建立微波辅助萃取-高效液相色谱法(MAE—HPLC)分析鱼腥草药材中槲皮素含量的方法。方法采用Diamonsil C18柱(5μm,150mm×4.6mm),以甲醇-0.2%三氟乙酸(50:50)为流动相,350rim处波长检测,流速1.0mL/min。微波辅助萃取条件的溶剂是10mL甲醇,提取功率为400w,提取时间为4min。结果槲皮素在0.01~0.2μg的范围内线性关系关系良好(r=0.9991),高、中、低3个浓度的平均加样回收率为98.32%~103.22%,RSD为1.21%~2.92%(n=3);样品的天内和天间RSD为0.42%和2.99%。结论该方法简便、准确,适用于分析测定鱼腥草中槲皮素的含量。Objective To develop an MAE- HPLC method for the determination of quercetin in Houttuynfa cordata Thunb. Methods Quercetin was determined by HPLC on Diamonsil C18 column (150 mm × 4. 6 mm,5 μm) at 30 ℃ with methanol-0.2% trifluoroacetic acid (50:50)as the mobile phase. Detection wavelength was 350 nm. The flow rate was 1.0 mL/min. Quercetin was extracted by MAE with 10 mL methanol using extraction condition of 400 w and 4 min. Results The standard curves for quercetin were linear over the range of 0.01 - 0.2 tLg (r = 0. 999 1). The average recoveries of quercetin were between 98.32% and 103.22% with RSD from 1.21% and 2.92%(n = 3). The RSD of intra-day and inter-day were 0.42 % and 2.99 %, respectively. Conclusions The method was simple and accurate. It is adapted to determine and analyze quercetin in Houttuynfa cordata Thunb.
