摘 要:黄瓜单性结实特性是其重要的经济性状,选育单性结实品种是现代黄瓜育种中重要的目标性状之一。本实验以黄瓜单性结实品系EP-6和非单性结实品系ZR-2为亲本构建群体,采用分离群体分组分析(bulkedsegregantanalysis,BSA)筛选与黄瓜单性结实基因连锁的分子标记。用65条ISSR引物进行PCR扩增筛选,其中引物N92在亲本和DNA池之间扩增出一条大小为850bp多态性片段,经F2代群体135个单株验证,该片段稳定出现,在单性结实植株中表现为无带,在非单性结实植株中表现为有带,可以作为鉴别单性结实与非单性结实的标记引物。遗传连锁分析表明,该标记与单性结实基因连锁距离为18.9cM,为黄瓜单性结实基因的分子标记筛选和分子标记辅助育种奠定了初步基础。Parthenocarpy is one of important economic traits and also is an important breeding objective in modern cucumber breeding program. The separate progenies from the cross between parthenocarpic line EP-6 and non-parthenocarpic line ZR-2 were employed to identify the molecular markers linked to the parthenocarpy of cucumber with the method of BSA (bulked segregation anlysis). A band of 850 bp was generated in the line of non-parthenocarpy by the primer N92 both in the parent and in the DNA pool. Accordingly, it is possible that the primer N92 might be used to phenotyping the trait of cucumber parthenocarpy, and further we calculated the genetic distance between the marker and cucumber parthenocarpy trait that might be 18.9 cM. In present research it would provide the preliminary basis for marker assisted selection in cucumber breeding program and gene tagging of cucumber parthenocarpy.
