摘 要:目的建立小型低压低氧处理系统,进行间歇性低氧心脏保护作用的研究。方法自行研制小型低压低氧处理系统,制备间歇性低氧适应大鼠模型。雄性成年Sprague-Dawlay(SD)大鼠12只,随机分为两组,对照组(CON)和间歇性低压低氧(intermittent hypobaric hypoxia,IHH)组。IHH组于低压氧舱接受相当于5000 m海拔的低压低氧(PO2=11.1%)处理28 d,6 h/d。CON组动物除不给予低压低氧外,其余处理同IHH组。定期测定动物体质量并观察动物的一般情况。应用Langendorff离体心脏灌流技术,给予心脏缺血(停灌30 min)/再灌注(复灌60 min)处理,记录心功能和冠脉流量变化。实验结束,取心测定心脏质量。结果①IHH组大鼠机体活动、体质量增长与CON组无明显差异。②IHH组大鼠的全心/体质量、左心室/体质量、右心室/体质量与CON组无明显差异。③心脏停灌缺血30 min后再灌注60 min,IHH组大鼠各项心功能指标的恢复均好于CON组。结论低压低氧系统处理大鼠的心脏具有明显抗缺血/再灌注损伤作用。Objective To establish a hypobaric hypoxic treating system that is used to study the protective effect of intermittent hypobaric hypoxia on rats hearts. Methods The small animal hypobaric hypoxic treating system was manufactured by ourselves and 12 adult male Sprague- Dawlay rats were randomly divided into two groups, control group (Con) and intermittent hypobaric hypoxia group(IHH). The IHH rats were exposed to a simulated 5 000 m highland hypoxia in hypobaric chamber for 28 d,6 h/d. The Con rats were kept in the same environment as IHH except hypoxia exposure. The body weight of rats was measured weekly. The isolated rats heart was perfused and treated with 30 min global ischemia followed by 60 min reperfusion in Langendorff apparatus. Parameters of cardiac function and discharge of coronary circulation were recorded before ischemia and after reperfusion. At the end of experiment,the heart was removed for measurement of weight. Results ① There was no significant difference in physical activity and the increment of body weight between the two groups. ② There was no significant difference in heart weight/body weight, left ventricular weight/body weight and right ventricular weight/body weight between the two groups. ③ The recovery of cardiac function after 60 min reperfusion in IHH rats was much better than that in Con. Conclusion Intermittent hypobaric hypoxia treatment can protect heart against ischemia/reperfusion injury.
