摘  要:目的探讨已酮可可碱(PTX)对全身炎症反应综合征(SIRS)大鼠核因子-κB(NF—κB)的调控。方法腹腔注射内毒素建立SIRS大鼠实验模型,随机分为正常对照组、SIRS组、PTX组,实验0、1、2、3、4及6h用酶联免疫吸附法(ELISA)检测血浆NF—κB活性及肿瘤坏死因子(TNF—α)、白介素-10(IL-10)含量水平。结果与正常对照组比较,SIRS组大鼠NF—κB活性和TNF-α、IL—10表达明显升高(P〈0.05);与SIRS组比较,PTX组大鼠NF—κB活性和TNF-α表达明显降低(P〈0.05);而IL-10表达明显升高(P〈0.05)。在PTX组,NF—κB的活性与TNF—α呈显著正相关(r=0.77,P〈0.05),与IL-10无明显相关性(r=-0.13,P〉0.05)。结论PTX可明显抑制SIRS大鼠的促炎症介质。其作用机制可能是抑制NF-κB的活性。Objective To explore the effects of peutoxifylline (FIX) on the activation of nuclear factor Kappa B (NF-κB) of rats with systemic inflammatory response syndrome. Methods The animal model of SIRS was established by endotoxin, 144 Wistar rats were random distributed into three groups, including control group SIRS group and PTX group, The activation of NF-κB and the expression of interleukin 10 (IL-10), tumor necrosis factor (TNF-α) were examined by ELISA at 0h, 1h ,2h, 3h, 4h and 6h respectively, Results Compared with control group, the activation of NF- κB and the expression of IL-10, TNF-α in SIRS group increased significantly ( P 〈0. 05). Compared with SIRS group, the activation of NF- κB and the expression of TNF - α in PTX group decreased significantly ( P 〈 0. 05), but the expression of IL-10 in FIX group increased significantly ( P 〈0. 05), In PTX group, there were significant positive correlation between the activation of NF-κB and the expression of TNF-α( P 〈0, 05), There are no correlation between the level of NF-κB and the expression of IL-10 ( P 〉 0. 05), Conclusion PTX may alleviate the inflammatory reaction of SIRS by inhibiting NF-κB activation.
