摘  要:目的:探讨定量缓释rbFGF/PLGA植入片对兔下颌牵张成骨的促进作用。方法:18只新西兰大白兔随机分为2组,每组9只,建立兔下颌双侧牵张成骨动物模型。将定量缓释rbFGF/PLGA植入片植入实验组牵张区两侧,对照组牵张区植入空白PLGA。牵张后固定期不同时间分别进行影像学、组织学检查,骨密度及生物力学检测,并测定ALP活性及骨钙素含量评估新生骨质量。结果:2组下颌骨牵张后间隙均有新骨形成,影像学及组织学结果显示实验组新骨形成速度和质量优于对照组,实验组骨密度及生物力学检测结果亦优于对照组(P<0.05)。结论:rbFGF/PLGA植入片中rbFGF生物活性保存良好,具有定量缓释作用,能明显促进兔下颌牵张成骨的速度和质量。Objective: To fabricate sustained release rbFGF/PLGA implant tablets and investigate their effects in accelerating bone growth during mandibular distraction osteogenesis (DO) in rabbits. Methods: 18 New Zealand white rabbits were divided randomly into two groups, 9 in each group. Bilateral mandibular DO models were established. The rbFGF/PLGA implant tablets were fabricated by solvent evaporation technique and were implanted into 9 rabbits in group A. PLGA implant tablets were implanted into group B as control. Normal distraction forces were applied to all mandibles of the rabbits. Radiographic and histological observation, biomechanical, ALP activity and osteocalcin concentration measurement were performed to evaluate the bone regeneration. Results:The rbFGF/PL- GA implant tablets showed sustained release effect. Bone regeneration was found in the distraction gap after mandibular lengthening in both groups. The formation rate of new bone and the density of the regenerated bone trabeculae of the rabbits treated by rbFGF/PLGA tablets were superior to those of the control group ( P 〈 0.05 ). Conclusion : The rbFGF/PLGA implant tablets have excellent pharmaceutical properties by sustained release. They can accelerate bone regeneration during mandibular DO in rabbits.
