摘 要:目的研究模拟失重对大鼠心肌组织中部分细胞因子基因表达谱的影响,探讨模拟失重条件下心脏结构/功能可能发生的变化机制。方法将成年雄性Wistar大鼠随机分为正常对照组(Con)及尾部悬吊模拟失重组(SUS),应用包含96种基因的细胞因子基因芯片检测各组细胞因子表达水平,选取部分表达变化细胞基因,应用Realtime-PCR给予验证。结果与Con组相比,模拟失重可导致心肌组织中17种细胞因子基因表达差异超出2倍,5种(IFNA4、IL-15、IL-1b、LT-b、FGF7)被上调,12种(IGF-1、IGF-II、FGF5、VEGF-D、VEGF-C、IFNr、IL-11、IL-12B、IL-13、IL-17、IL-18、CD40L)被下调。其中,生长因子呈现较普遍下调趋势,炎性类细胞因子表达水平变化不一。结论模拟失重2wk可导致心肌组织中细胞因子网络调控平衡偏移,促生长性、保护性细胞因子普遍下调,而致病性、炎性细胞因子部分明显上调,其可能是失重/模拟失重条件下心肌组织功能结构重塑的重要调控机制。Objective To investigate the effect of simulated microgravity on partial cytokine gene expression spectrum in rat myocardium and to explore the possible mechanism of cardiac function and structural changes under simulated microgravity, Methods Male Wistar rats were randomly devided into either simulated microgravity, tail-suspended group (SUS) or control group (Con). The cytokines gene expression levels in rat myocardium were detected with 96 kind cytokine gene chips. Partial of the results were determined with Real time-PCR, Results Comparison with Con group, 17 of 96 cytokine genes of SUS group were changed after simulated microgravoty, Five of them( IF- NA4, IL-15, IL-1b, LT-b, FGF7) were regulated upward and 12 (IGF-1, IGF-2, FGF5, VEGF -D, VEGF-C, IFN r, IL-Ⅱ, IL-12B, IL-13, IL-17, IL-18, CD40L) were regulated downward, Expression of most growth factors presented a down trend, but the expression levels of the inflammatory cytokines had different trends. Conclusion It is suggested that 2 wk simulated microgravity can induce expression changes of many cytokine genes and induced unbalanced regulation in the cytokine net of rat myocardium, The promoting growth and protecting cytokines are generally regulated downward, but parts of pathogenic and inflammatory cytokines are obviously regulated upward, It is showed that cytokines gene expression may play an important role in the cardiac remodeling mechanism under weightlessness.
