摘 要:目的:寻找缺血性脑血管疾病的特异蛋白质并探讨针刺对其差异蛋白质表达的影响。方法:用固相pH梯度双向凝胶电泳分离正常组、缺血性脑血管病及针刺组大鼠的总蛋白质,经考马斯亮兰染色,PDQuest 7.0软件分析。结果:与正常组匹配,缺血性脑血管病组与针刺组大鼠脑皮质图象匹配率分别为82%、85%,且重复性较好,其蛋白质主要分布在等电点pI4-8,分子量Mr20-75 kD范围。与正常组比较,缺血性脑血管病组中蛋白质点表达上升2倍及以上的点有9个,下降2倍及以上的点有11个。而在针刺组中这些表达上升的点均有下降,有10个点下降达2倍以上,其中有6个下降后接近于正常组;这些表达下降的点均有上升,有7个点上升达2倍以上,其中有4个上升后接近于正常组。结论:上述有显著差异的蛋白质可能是缺血性脑血管疾病的相关蛋白;针刺改善缺血性脑血管疾病的临床症状也可能与上述蛋白质的改变有关。Objective: To explore the characteristic proteins of ischemic cerebrovascular disease and discuss the influence of acupuncture on discrepancy protein expression. Methods: The total proteins of pallium of normal control rats, the rats with ischemic cere-brovasenlar disease and the aeupuneturing rats were separated by immobilized pH gradient - based two - dimensional gel deetrophoresis (2-DE). After Coomassie Brilliant Blue staining, PDQuest 7.0 software was applied to analyse 2-DE images. Results: Matching with the normal group, the average matching rates of the proteins of isehemle eerebrovasenlar disease group and acupuncture group were respectively 82% and 85% with good reproduelbility. Proteins were mainly displayed at the range of pI 4-8, Mr 20-75 kD. In the group with isehemie eerebrovascular disease, there were nine proteins whose expression had increased at least much twice than in the normal group while 11 proteins whose expression had decreased at least less than half of those in the normal group. After being aeupunctured, the excessively expressed proteins decreased, 10 of them decreased at least less than half of those in the isehemie eerebro-vascular disease group and six of them were dose to those in the normal group. While the depressed proteins increased, seven of them increased at least much twice than those in the isehemic eerebrovasenlar disease group and four of them were dose to those in the normal group. Condusion: These proteins that have significant deviation maybe are GAP -associated proteins of isehemie eerebrovasenlar disease. The reason why the aeupuneure can improve the clinical symptom of isehemie cerebrovascular disease may have some relations with the changes of proteins we talked above.
