天麻钩藤饮对高血压肝阳上亢证大鼠肾上腺蛋白质的影响  

Effect of Tianmagouteng Decoction on Proteins of Adrenal Glands in Hypertensive Rats with Hyperactivity of Liver-YANG Syndrome

摘　　要：目的:从蛋白质组角度探讨高血压肝阳上亢证证候相关蛋白及天麻钩藤饮降血压和改善肝阳上亢证症状的机理。方法:用固相pH梯度双向凝胶电泳分离正常大鼠以及肝阳上亢证大鼠天麻钩藤饮治疗前后肾上腺的总蛋白质,经考马斯亮兰染色,PDQuest 7.0软件分析。结果:与正常组匹配,肝阳上亢证组与治疗组大鼠肾上腺图象匹配率分别为90%、95%,且重复性较好,其蛋白质主要分布在等电点PI 3-8,分子量Mr 14.4-75 kD范围。与正常组比较,肝阳上亢证组中蛋白质点表达上升2倍及以上的点有22个,下降2倍及以上的点有15个。而在治疗组中这些表达上升的点均有下降,有14个点下降达2倍以上,其中有10个下降后接近于正常组;这些表达下降的点均有上升,有9个点上升达2倍以上,其中有5个上升后接近于正常组。结论:这些有显著差异的蛋白质可能是高血压肝阳上亢证证候相关蛋白,天麻钩藤饮的降压和改善症状也可能与上述蛋白质的改变有关。Objective：From proteome to investigate the protein associated Hyperactivity of liver-YANG syndrome （HLYS）, and discuss the mechanism of decreasing blood pressure of the Tianmagouteng Decoction on HLYS of hypertensive rats. Metheds.The total proteins of adrenal glands in normal control rats and HLYS of hypertensive rats were seperated by immobilized pH gradient - based two-dimensional gel eleetrophoresis（2-DE）, After eoomassie blue staining, PDQuest 7.0 software was applied to analyze 2-DE images. Results ： Compared with normal control group, the average matching rates of adrenal glands of HLYS group and treatment group were respectively 90%, 95% with reproducibility. Proteins were mainly displayed at pI 3-8, Mr14.4 -75kD range. At HLYS rats , 22 spots expressed two times than those of normal rats, 15 spots expressed half than those of normal rats. After treatment, 14 spots of excessively expressed proteins decreased, 10 spots of them were near to those of normal rats. 9 spots of depressed proteins increased, 5 spots of them were near to those of normal rats. Conclusions. These protein spots with significant difference may be associated with Hyperactivity of HLYS in essential hypertension. After theraping by drinking tianmagouteng decotioin, the blood pressure of HLYS of hypertensive rats decreased, it may be related with some of these protein spots.
