摘  要:目的建立中枢神经系统脱髓鞘疾病蛋白质组学技术体系,寻找并鉴定与中枢神经系统脱髓鞘疾病发生有关的生物标识物。方法以中枢神经系统脱髓鞘疾病患者的脑脊液为研究对象,分别采用固相pH梯度双向凝胶电泳分离总蛋白质,凝胶经银染显色后,用图像分析软件进行比较分析、识别差异表达的蛋白质,离线应用基质辅助激光解析电离飞行时间质谱得到相应的肽指纹图谱;同时用超高压液相色谱分离酶解肽段,在线采用电喷雾四极杆飞行时间质谱鉴定肽段氨基酸序列;搜索数据库鉴定部分蛋白质。结果分别获得中枢神经系统脱髓鞘疾病患者组和对照组的脑脊液双向电泳蛋白质组表达图谱,筛选并鉴定出表达有明显差异的蛋白质10个,其中在疾病组中血红蛋白有明显上调而前列腺素H2表达下调明显。结论这些差异蛋白可能成为具有临床诊断意义的中枢神经系统脱髓鞘疾病潜在标识物,从而为中枢神经系统脱髓鞘疾病的早期诊断和治疗提供理论和实验依据。Objective To establish the comparative proteomic technology platform for Delnyelinative disease of the central nervous system in cerebrospinal fluid and to screen out disease related protein markers by proteome analysis. Methods Proteins and peptides in CSF were separated by two-dimensional gel electrophoresis(2DE) and ultra-performance liquid chromatographer( UPLC) and identified by matrix assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) and electrospray ionization(ESI-MS) to study differential proteomic expression in the cerebrospinal fluid(CSF) between patients with Demy- elinative disease (the experimental group) and with other neurological diseases (ONDs) (the control group). Results Different 2-DE maps were obtained for the protein spats in the experimental group and the control group gels. Image analysis of silverstained 2-DE gels revealed that 10 protein spots showed a significantly differential expression between the two groups of CSF sampies. After searching the database, it was found that in the CSF of Demyelinative patients, the expression of hemoglobin was increased. However, the expression of Prostaglandin-H2 Disomerase was decreased. Conclusion These identified proteins have potential relations to Demyelinative disease physiology. It would be helpful to find new molecular markers for early diagnosis and treatment of the disease.
