摘  要:目的:观察荔枝核总皂苷体外抗乙型肝炎病毒(HBV)的作用.方法:以HepG2.2.15细胞为靶细胞,分别用不同浓度的荔枝核总皂苷和拉米夫定培养液培养,采用ELISA法及荧光定量PCR法测定细胞培养上清HBsAg,HBeAg及细胞外HBVDNA含量,评价荔枝核总皂苷对体外HBV的抑制作用.结果:荔枝核总皂苷对体外培养HepG2.2.15细胞分泌的HBsAg,HBeAg及细胞外HBVDNA的50%抑制浓度(IC50)分别为0.096,0.085和0.128g/L,治疗指数(TI)分别为6.98,7.88和5.23.0.25g/L拉米夫定组和0.4g/L荔枝核总皂苷组在d3,d6对体外培养HepG2.2.15细胞分泌的HB-sAg,HBeAg及细胞外HBVDNA的抑制作用差异无统计学意义(P>0.05).结论:荔枝核总皂苷具有一定的体外抗HBV作用.AIM: To evaluate the inhibitory effects of the total saponin extracted from the semen litchi against hepatitis B virus (HBV) in vitro. METHODS : HepG2.2. 15 cells, as target cells, were exposed to different concentrations of total saponin and Lamivudine respectively. Enzyme-linked immunosorbent assay (ELISA) was used to assess HBsAg and HBeAg; fluorescent quantitative PCR was applied to quantify extracellular HBV DNA. RESULTS: The 50% inhibitory concentration (ICso) of total saponin against HBsAg, HBeAg and extracellular HBV DNA was 0. 096, 0. 085 and 0. 128 g/L respectively, and treatment index (TI) was 6.98, 7.88, 5.23 g/L respectively. The inhibitory effects of 0.25 g/L Lamivudine and 0.4 g/L total saponin against HBV had no significant difference between day 3 and day 6 (P 〉 0.05). CONCLUSION : The total saponin extracted from semen litchi inhibits HBV replication in vitro to some extent.
