摘 要:目的:在基因转录水平了解细胞骨架相关基因在大鼠肝再生中的作用.方法:采用搜集网站资料和查阅相关论文等方法获得上述基因,用Rat Genome 230 2.0芯片检测它们在大鼠再生肝中的表达情况,用真、假手术比较方法确定肝再生相关基因.结果:初步证实249个参与细胞骨架结构和功能的基因与肝再生相关.其中,肝再生启动[部分肝切除(PH)后0.5~4h]、G0/G1过渡(PH后4~6h)、细胞增殖(PH后6~66h)、细胞分化和组织结构功能重建(PH后72~168h)等4个阶段起始表达的基因数为107,29,137和5,基因总表达的次数为209,113,1063和326.表明相关基因主要在肝再生启动阶段起始表达,在不同阶段发挥作用.它们表达的相似性分为均上调、上调占优势、均下调、下调占优势、上调和下调相近等5类,涉及107,37,67,28和10个基因,它们共上调1118次,下调480次,分为40种表达模式.表明肝再生中细胞生理生化活动具有阶段性、多样性和复杂性.结论:肝再生早期和前期微丝形成相关基因表达增强,早期、前期和后期微管形成相关基因表达增强,早期、中期和后期中间纤维形成相关基因表达增强,前期核纤层形成相关基因表达增强.AIM: To study the roles of the cyteskeleton-associated genes in liver regeneration (LR) at the transcriptional level. METHODS: The above genes were obtained by collecting data from the databases and the related thesis, and their expression profiles in rat regenerating liver were detected by Rat Genome 230 2.0 array. The LR-associated genes were identified by the comparison of the gene expression difference between sham operation group and partial hepatectomy (PH) group. RESULTS: 249 cyteskeleton-associated genes were proved to be LR-associated. The initially and totally expressed number of these genes occurring in initial phase of LR ( 0.5- 4 h after PH ), G0/G1 transition (4-6 h after PH), cell proliferation (6 -66 h after PH), cell differentiation and structure-functional reconstruction (72 -168 h after PH) were 107, 29, 137, 5 and 209, 113, 1063, 326, respectively, illustrating that the associated-genes initiated their expression mainly in the early phase, and worked in the different phases. Their expression similarity was classified into 5 groups ineluding only up-, predominantly up-, only down-, predominantly down-, and approximately up/down-regulated, involving 107, 37, 67, 28 and 10 genes, respectively; the frequencies of upexpression and down-expression were 1118 and 480 ; their expression patterns were categorized into 40 types, showing that the physiological and biochemical activities in LR were staged, diverse and intricate. CONCLUSION: Microfilament-associated genes at forepart and prophase, microtubule-associated genes at forepart, prophase and anaphase, intermediatefilament-associated genes at forepart, metaphase and anaphase, lamin-associated genes at prophase are all increased at the mRNA level.
