人白介素17基因的分离及其在大肠杆菌中的表达

摘　　要：用ＲＴ－ＰＣＲ方法从活化的人外周血淋巴细胞中分离了人白介素１７（ｈＩＬ－１７）基因，序列分析结果表明与文献报道的完全相符。以质粒ｐＢＶ２２０为表达载体，在大肠杆菌中高效表达了ｈＩＬ－１７基因，重组蛋白以包涵体的形式存在，约占菌体总蛋白的量４０％。Human interleukin 17 gene was isolated from activated human peripheral blood lymphoid cells by RT PCR. The gene was sequenced and conformed with that reported. hIL 17 gene was cloned into the plasmid pBV220 and expressed in E.coli in the form of inclusion bodies. The recombinant protein amounted to 40% of total proteins in bacterial cells.
