摘 要:目的建立用于测定肌萎缩侧索硬化(ALS)患者脑脊液中氨基酸类神经递质的方法,观察患者用肌萎灵注射液治疗前后脑脊液中氨基酸类神经递质的变化。方法采集42例患者的脑脊液,用邻苯二甲醛(OPA)柱前衍生变增益HPLC法测定脑脊液中天冬氨酸、谷氨酸、甘氨酸、γ-氨基丁酸的含量。结果4种氨基酸在20min内分离度、线性关系均良好;42例ALS患者治疗后与治疗前相比,脑脊液中兴奋性的天门冬氨酸和谷氨酸水平明显降低,而抑制性的甘氨酸和γ-氨基丁酸水平变化不明显。结论本方法快速、准确、灵敏度高,适用于脑脊液中神经递质类氨基酸的含量测定。Objective To establish a method for determination the contents of amino acid-like neurotransmitter in cerebrospinal fluid (CSF) of amyotrophic lateral sclerosis (ALS) patients. The changes of amino acid - like in CSF were observed in the patients before and after the treatment with Jiweiling Injection. Method The CSF were collected from 42 patients, in which the contents of aspartic acid, glutamic acid, glycine and γ-Aminobuyric acid were detected with HPLC of OPA pre-column derivation. Result Above four kinds of amino acid were separated well with a good linear relationship within 20 minutes. After the treatment in 42 patients the contents of aspartic acid and glutamic acid were decreased significantly, while those of glycine and γ-Aminobuyric acid did not change. Conclusion The method can be performed rapidly and accurately with a higher sensitivity. It can be used in the quantitative determination of amino acid-like neurotransmitter in CSF.
